Introduction
NICD is the product of 3 proteolytic cleavages of NOTCH1, 1st by furin at S1, then by ADAM17 at S2 (NEXT), then by gamma-secretase at S3. NEXT cleavage occurs between gly1743 & val1744 of parent Notch-1 (S3). Cleavage serves to release the NOTCH1 intracellular domain (NICD) from the membrane. NICD then translocates to the nucleus, where it functions as a transcriptional activator in concert with CSL family members.
