• may protect lysosomal membrane from autodigestion
• maintenance of the acidic environment of the lysosome,
• adhesion when expressed on the plasma membrane
• inter- & intracellular signal transduction
• may act as receptor for proteins to be imported into the lysosome

• O- & N-glycosylated
• some of the 16 N-linked glycans are polylactosaminoglycans
• belongs to the LAMP family

Compartment

• cell membrane, endosome membrane, lysosome membrane
• shuttles between lysosomes, endosomes, & the plasma membrane

Named isoforms=2: LAMP-2A, LAMP-2B

• lysosomes
• degranulated platelets
• activated neutrophils (weak expression)
• endothelium (weak expression)
• LAMP-2A:
  • high expression in placenta, lung, liver
  • less expression in kidney and pancreas
  • low expression in brain and skeletal muscle
• LAMP-2B:
  • high expression in skeletal muscle
  • less expression in brain, placenta, lung, kidney and pancreas
  • low expression in liver

Pathology

• mutations associated with glycogen storage disease IIb
• may play a role in tumor metastasis
