Function
- activates ubiquitin by first adenylating with ATP its carboxy-terminal Gly & thereafter linking this Gly to the side chain of Cys in E1, yielding an ubiquitin-E1 thioester & free AMP
- protein degradation; protein ubiquitinylation
- binds & is involved in the conjugation of G1P2/ISG15
Structure
- monomer (putative)
- belongs to the ubiquitin-activating E1 family
- there are 2 active sites within the E1 molecule, allowing it to accommodate 2 ubiquitin moieties at a time, with a new ubiquitin forming an adenylate intermediate as the previous one is transferred to the thiol site
Expression
- expressed in a variety of normal & tumor cell types
Pathology
- expression is diminished in lung cancer cell lines
