Function
- pyrimidine base degradation
- catalyzes reduction of uracil & thymine to 5,6-dihydrouracil & 5,6-dihydrothymine
- degradation of chemotherapeutic drug 5-fluorouracil
- amino-acid biosynthesis, beta-alanine biosynthesis
Cofactor: binds 2 FAD, 2 FMN, 2 4Fe-4S clusters
Structure
- homodimer
- belongs to the dihydropyrimidine dehydrogenase family
- contains 3 4Fe-4S ferredoxin-type domains
Compartment
Expression
- expressed in most tissues
- greatest expression in liver & peripheral blood mononuclear cells
Pathology
- defects in DPYD are the cause of dihydropyrimidine dehydrogenase deficiency
