Function

• phosphorylation of Ins(1,4,5)P3 to yield Ins(1,3,4,5)P4
• also phosphorylates Ins(2,4,5)P3 to yield Ins(2,3,4,5)P4
• activated by Ca+2/calmodulin
• inhibited by high concentrations of substrate Ins(1,2,4)P3,
• allosteric activation by the product Ins(1,3,4,5)P4

Compartment

• shuttles actively between nucleus & cytoplasm
• associated with both import & export activity

Expression

• expressed in pancreas, skeletal muscle, liver, placenta > kidney, brain

Pathology

• defects in ITPKC are a cause of Kawasaki disease
