Function
- role in physiological regulation of intracellular CoA (putative)
- regulated by feedback inhibition by CoA & its thioesters
- strongly inhibited by acetyl-CoA & by manyl-CoA & also inhibited by high concentration of non-esterified CoA (CoASH)
- cofactor biosynthesis, coenzyme A biosynthesis
- coenzyme A from pantothenate: step 1/5
ATP + (R)-pantothenate <--> ADP + (R)-4'-phosphopantothenate
Structure
- N-terminal extension, present in isoform 1 may be the regulatory domain
- belongs to the type 2 pantothenate kinase family

Expression
- isoform 1 is expressed at high levels in brain, heart, kidney, liver, skeletal muscle & testis
- isoform 2 is detected at much lower levels in kidney, liver, brain & testis & is not detected in heart or skeletal muscle
- isoform 1 is induced by bezafibrate, a hypolipidemic drug which acts as an agonist of PPARA; isoform 2 levels decrease slightly
