• method of DNA sequencing based upon sequencing by synthesis
• relies on release of pyrophosphate upon nucleotide incorporation
• the DNA sequence is determined by light emitted upon incorporation of complementary nucleotide since only one of the four nucleotides is available at a time for incorporation on the single-stranded template
