Function

  • catalyzes degradation of compounds including: putrescine, histamine, spermine, & spermidine, substances involved in allergic & immune responses, cell proliferation, tissue differentiation, tumor formation, & possibly apoptosis
  • placental DAO is thought to play a role in the regulation of female reproductive function
  • topaquinone is generated by copper-dependent autoxidation of a specific Tyr
  • inhibited by amiloride (competitive inhibition)
RCH(2)NH(2) + H2O + O2 <--> RCHO + NH3 + H2O2

Cofactor:

  • binds 1 copper ion per subunit (putative)
  • binds 2 Ca+2 per subunit (putative)
  • contains 1 topaquinone per subunit (putative)

Structure

belongs to the copper/topaquinone oxidase family
