Function
- lipid kinase
- role in lipid metabolism, glycerolipid metabolism
- can phosphorylate both monoacylglycerol & diacylglycerol to form lysophosphatidic acid & phosphatidic acid, respectively
- does not phosphorylate sphingosine
- overexpression increases formation & secretion of lysophosphatidic acid, resulting in transactivation of EGFR & activation of the downstream MAPK signaling, leading to increased cell growth
ATP + acylglycerol <--> ADP + acyl-sn-glycerol 3-phosphate
ATP + 1,2-diacylglycerol <--> ADP + 1,2-diacyl- sn-glycerol 3-phosphate
Cofactor: Mg+2
Structure
contains 1 DAGKc domain
Compartment
Alternative splicing
named isoforms=2
Expression
- highly expressed in muscle, heart, kidney & brain
Pathology
- overexpressed in prostate cancer
